RESUMO

The plant-specific NAC gene family is one of the largest transcription factor families, participating in plant growth regulation and stress response. Despite extensive characterization in various plants, our knowledge of the NAC family in oat is lacking. Herein, we identified 333 NAC genes from the latest release of the common oat genome. We provide a comprehensive overview of the oat NAC gene family, covering gene structure, chromosomal localization, phylogenetic characteristics, conserved motif compositions, and gene duplications. AsNAC gene expression in different tissues and the response to various abiotic stresses were characterized using RT-qPCR. The main driver of oat NAC gene family expansion was identified as segmental duplication using collinearity analysis. In addition, the functions of AsNAC109 in regulating abiotic stress tolerance in Arabidopsis were clarified. This is the first genome-wide investigation of the NAC gene family in cultivated oat, which provided a unique resource for subsequent research to elucidate the mechanisms responsible for oat stress tolerance and provides valuable clues for the improvement of stress resistance in cultivated oat.

Prostate cancer (PCa) is a common malignancy in elderly men. We have applied Traditional Chinese Medicine CFF-1 in clinical treatments for PCa for several years. Here, we aimed to identify the underlying mechanism of CFF-1 on PCa using network pharmacology and experimental validation. Active ingredients, potential targets of CFF-1 were acquired from the public databases. Subsequently, protein-protein interaction (PPI) and the herbs-active ingredients-target network was constructed. A prognostic model for PCa was also constructed based on key targets. In vitro experiments using PCa cell lines CWR22Rv1 and PC-3 were carried out to validate the potential mechanism of CFF-1 on PCa. A total of 112 bioactive compounds and 359 key targets were screened from public databases. PPI and herbs-active ingredients-target network analysis determined 12 genes as the main targets of CFF-1 on PCa. Molecular docking studies indicated that the primary active ingredients of CFF-1 possess strong binding affinity to the top five hub targets. DNMT3B, RXRB and HPRT1 were found to be involved in immune regulation of PCa. In vitro, CFF-1 was found to inhibit PCa cell proliferation, migration, invasion and induce apoptosis via PI3K-Akt, HIF-1, TNF, EGFR-TKI resistance and PD-1 checkpoint signaling pathways. This study comprehensively elucidates the underlying molecular mechanism of CFF-1 against PCa, offering a strong rationale for clinical application of CFF-1 in PCa treatment.

Hemicelluloses constitute approximately one-third of the plant cell wall and can be used as a dietary fiber and food additive, and as raw materials for biofuels. Although genes involved in hemicelluloses synthesis have been investigated in some model plants, no comprehensive analysis has been conducted in common oat at present. In this study, we identified and systematically analyzed the cellulose synthase-like gene (Csl) family members in common oat and investigated them using various bioinformatics tools. The results showed that there are 76 members of the oat Csl gene family distributed on 17 chromosomes, and phylogenetic analysis indicated that the 76 Csl genes belong to the CslA, CslC, CslD, CslE, CslF, CslH, and CslJ subfamilies. A total of 14 classes of cis-acting elements were identified in the promoter regions, including hormone response, light response, cell development, and defense stress elements. The collinearity analysis identified 28 pairs of segmentally duplicated genes, most of which were found on chromosomes 2D and 6A. Expression pattern analysis showed that oat Csl genes display strong tissue-specific expression; of the 76 Csl genes, 33 were significantly up-regulated in stems and 30 were up-regulated in immature seeds. The expression of most members of the AsCsl gene family is repressed by abiotic stress, while the expression of some members is up-regulated by light. Immunoelectron microscopy shows that the product of AsCsl61, a member of CslF subfamily, mediates (1,3; 1,4)-ß-D-glucan synthesis in transgenic Arabidopsis. These findings provide a fundamental understanding of the structural, functional, and evolutionary features of the oat Csl genes and may contribute to our general understanding of hemicellulose biosynthesis. Moreover, this information will be helpful in designing experiments for genetic manipulation of mixed-linkage glucan (MLG) synthesis with the goal of quality improvement in oat.

We aimed to explore the predictive value of abnormal electrocardiogram (ECG) changes in non-small-cell lung carcinoma non-small cell lung cancer (NSCLC) patients for near-term mortality. Seventy-eight NSCLC patients hospitalized in Chaohu Hospital Affiliated to Anhui Medical University from October 2017 to August 2021 were selected as the observation group. Baseline medical history and type of ECG abnormalities were the same. Seventy-eight patients were included in the control group. To compare the difference in mortality within 3 months between the observation group and the control group after the occurrence of the same type of electrocardiogram abnormalities, and to explore the value of electrocardiogram abnormalities in the early warning of NSCLC patients recent death. When the average ventricular rate of patients in the observation group was ≥100 beats/minute, ST-segment depression ≥0.05 mm, QRS voltage decrease ≥0.5 mm, new atrial fibrillation and other electrocardiogram changes, all patients died within 3 months. The control group with the same abnormal ECG had no death within 3 months (P < .05). In the resting 12-lead synchronous ECG in NSCLC patients, when the average ventricular rate ≥100 beats/minute, QRS wave voltage significantly decreased ≥0.5 mm, ST segment depression ≥0.05 mm, new atrial fibrillation appeared, it suggested that the patients' cardiac function decreased and the potential for near-term adverse outcomes is high.

Aspergillosis is a rising concern worldwide; however, its prevalence is not well documented in China. This retrospective study determined Aspergillus's epidemiology and antifungal susceptibilities at Meizhou People's Hospital, South China. From 2017 to 2022, the demographic, clinical, and laboratory data about aspergillosis were collected from the hospital's records and analysed using descriptive statistics, chi-square test, and ANOVA. Of 474 aspergillosis cases, A. fumigatus (75.32%) was the most common, followed by A. niger (9.92%), A. flavus (8.86%), and A. terreus (5.91%). A 5.94-fold increase in aspergillosis occurred during the study duration, with the highest cases reported from the intensive care unit (52.74%) - chronic pulmonary aspergillosis (79.1%) and isolated from sputum (62.93%). Only 38 (8.02%) patients used immunosuppressant drugs, while gastroenteritis (5.7%), haematologic malignancy (4.22%), and cardiovascular disease (4.22%) were the most prevalent underlying illnesses. In A. fumigatus, the wild-type (WT) isolates against amphotericin B (99.1%) were higher than triazoles (97-98%), whereas, in non-fumigatus Aspergillus species, the triazole (95-100%) WT proportion was greater than amphotericin B (91-95%). Additionally, there were significantly fewer WT A. fumigatus isolates for itraconazole and posaconazole in outpatients than inpatients. These findings may aid in better understanding and management of aspergillosis in the region.

Objective: Cryptococcosis is a fatal infection that can affect both immunocompetent and immunocompromised patients, and it is little understood in China's various regions. This research aimed to look at the epidemiology, risk factors, and antifungal susceptibility pattern of Cryptococcus neoformans in eastern Guangdong, China. Methods: A six-year (2016-2022) retrospective study was conducted at Meizhou People's Hospital, China. Demographical, clinical, and laboratory data of cryptococcal patients were collected from hospital records and statistically analyzed using the chi-square and ANOVA tests. Results: Overall, 170 cryptococcal infections were recorded, of which meningitis accounted for 78 (45.88%), cryptococcemia for 50 (29.41%), and pneumonia for 42 (24.7%). The number of cases increased 8-fold during the study duration. The median age of patients was 58 years (Inter quartile range: 47-66), and the high proportion of cases was from the male population (n = 121, 71.17%). The underlying diseases were identified only in 60 (35.29%) patients, of which 26 (15.29%) were severely immunocompromised, and 26 (15.29%) others were mildly immunocompromised. A statistically significant difference was reported for chronic renal failure, and anemia (p < 0.05) persisted in cases of three infection types. A high number of non-wild type (NWT) isolates were found against amphotericin B (n=13/145, 8.96%), followed by itraconazole (n=7/136, 5.15%) and voriconazole (n=4/158, 2.53%). Only six isolates (3.79%) were multidrug-resistant, four of which were from cryptococcemia patients. Compared to meningitis and pneumonia, cryptococcemia revealed a higher percentage of NWT isolates (p < 0.05). Conclusion: In high-risk populations, cryptococcal infections require ongoing monitoring and management.

Candidiasis is a life-threatening disease that increases mortality in critically ill patients. However, such epidemiological data are still lacking in underdeveloped regions of China. A retrospective analysis (2016 to 2021) was conducted in Meizhou People's Hospital, China to study the burden of candidiasis, particularly candidemia, and antifungal susceptibilities of the species among hospitalized patients. Of the 7,864 candidiasis cases, 461 (5.86%) were candidemia cases. Candida albicans (64.25%) was the most identified species, followed by C. tropicalis (12.61%), C. glabrata (10.79%), and C. parapsilosis (9.79%). In non-C. albicans (NCA) candidemia cases, the number of C. glabrata cases was higher (102/461, 22.37%) than C. tropicalis (64/461, 14.04%). Gastrointestinal pathology, respiratory dysfunctions, septic shock, and malignancies were common underlying comorbidities, respectively. A central venous catheter was an independent risk factor for both C. albicans and NCA candidemia. The mortality rate was not statistically significant for either C. albicans or NCA. Amphotericin B and 5-flucytosine were highly effective (98 to 100%), while azoles were least effective (67.74 to 95.66%). Candidemia cases caused by C. tropicalis and C. glabrata had significantly lower azole susceptibility than non-candidemia-causing isolates. This study provides valuable information for prescribers to choose the right empirical therapy, for researchers to explore different resistance mechanisms, and for health care managers to control candidiasis better. IMPORTANCE This study provides important information on the burden of candidiasis, particularly candidemia, and the antifungal susceptibility of Candida species among hospitalized patients in an underdeveloped region of China. First, the finding that azoles were least effective against Candida species causing candidemia is particularly noteworthy, as it suggests the possibility of resistance to this class of antifungal agents. This information can guide the choice of empirical therapy and help in the selection of appropriate antifungal agents for the treatment of candidemia, thereby reducing the risk of resistance development. Second, the study provides important information for researchers to explore different resistance mechanisms in Candida species. Finally, the study has important implications for health care managers in controlling the spread of candidiasis. The high prevalence of candidemia cases in the study highlights the need for appropriate infection control measures to prevent the spread of the disease.

Objective: To study the effect of health education combined with personalized psychological nursing intervention on pregnancy outcome of pregnant women with gestational diabetes mellitus (GDM). Methods: 170 patients with GDM admitted to Guangdong Women and Children Hospital from January 2018 to December 2018 were selected as study subjects and randomly divided into two groups. During the period from diagnosis of GDM to termination of pregnancy, both groups were given routine education and routine examination, and the intervention group adopted health education combined with personalized psychological nursing interventions during pregnancy. The pregnancy weight, blood glucose index, compliance, disease awareness, self-adjustment management ability, satisfaction, and pregnancy outcome were measured before and after the intervention. Results: There were no statistically significant differences in pregnancy weight, fasting plasma glucose, and 2 h postprandial blood glucose between the two groups before intervention (P = 0.768, 0.605, and 0.762). After intervention, lower levels of the above indicators were obtained in the intervention group than in the control group (P < 0.001). The compliance and satisfaction with the intervention in the intervention group were significantly higher than those in the control group (P < 0.001). The intervention group had remarkably higher disease awareness rate and self-psychological adjustment and management ability than the control group (P < 0.001). Better pregnancy outcomes were observed in the intervention group compared with the control group (P < 0.001). Conclusion: For patients with GDM, health education combined with personalized psychological nursing on the basis of the conventional nursing can effectively control patients' condition and ensure a better pregnancy outcome, which merits widespread promotion.

The aim of this study was to prepare a 20(S)-protopanaxadiol nanocrystalline suspension and enhance the bioavailability of 20(S)-protopanaxadiol by intramuscular injection. 20(S)-Protopanaxadiol nanocrystalline suspension was prepared using an anti-solvent combined with ultrasonic approach, in which meglumine and bovine serum albumin were screened as the optimized stabilizer and the coating agent during spray drying process, respectively. The optimal nanocrystallines were nearly spherical with a uniform particle size distribution, the mean particle size, polydispersity index, and drug loading of which were 151.20 ± 2.54 nm, 0.11 ± 0.01, and 47.15% (w/w), respectively. Sterile 20(S)-protopanaxadiol nanocrystalline suspension was obtained by passing through a 0.22-µm membrane, and the average filtration efficiency (FE%) was 99.96%. The cumulative release percentage of 20(S)-protopanaxadiol nanocrystalline suspension was 92.36% 20(S)-protopanaxadiol within 60 min in vitro, which was relatively rapid compared with that of the physical mixture for 12.51% and the 20(S)-protopanaxadiol bulk powder for 9.71% during the same time interval. The sterile 20(S)-protopanaxadiol nanocrystalline suspension caused minimal irritation responses by histological examination, indicating a good biocompatibility between the 20(S)-protopanaxadiol nanocrystalline suspension and muscle tissues. In pharmacokinetic study, the absolute bioavailability of 20(S)-protopanaxadiol nanocrystalline suspension for intramuscular injection and for oral gavage was 5.99 and 0.03, respectively. In summary, the 20(S)-protopanaxadiol nanocrystalline via intramuscular injection is an efficient drug delivery system to improve its bioavailability.

A 12-week feeding trial was conducted to investigate the effect of dietary daidzein on the intestinal mucosal barrier function and the intestinal microbiota profile of juvenile turbot (Scophthalmus maximus L.). Three isonitrogenous and isolipidic experimental diets were formulated to contain 0 (FM), 40 (D.40) and 400 (D.400) mg kg-1 daidzein, respectively. Fish fed D.400 had significantly lower growth performance than fish fed D.40. Dietary daidzein significantly increased the feed efficiency, while significantly decreased the feed intake. Daidzein supplementation increased the activity of total anti-oxidative capacity and the gene expression of anti-inflammatory cytokine transforming growth factor-ß1, Mucin-2 and tight junction proteins (Tricellulin, Zonula occludens-1 transcript variant 1, Zonula occludens-1 transcript variant 2 and Claudin-like and Occludin), and down-regulated the gene expression of pro-inflammatory cytokines interleukin-1ß and tumor necrosis factor-α in the intestine of turbot. Dietary daidzein increased intestinal microbial diversities, the abundance of several short chain fatty acids producers, and decreased the abundance of some potential pathogenic bacteria. However, D.400 had dual effects on lactic acid bacteria and increased the abundance of potential harmful bacterium Prevotella copri. Collectively, dietary daidzein at the levels of 40 and 400 mg kg-1 could enhance the intestinal mucosal barrier function and alter the intestinal microbiota of turbot. However, high dose of daidzein must be treated with caution for its unclear effects on intestinal microbiota of turbot in the present study.

The spatial distribution of the collectors in dust scrubber is key in determining the effectiveness of the dust removal process. In the present study, a high-speed camera was used to capture images of the distribution of the collectors. Some of the image information was extracted by image processing, such as the gray mean (GM), the angular second moment (ASM), and the entropy (ENT) from the gray-level co-occurrence matrix of the image. Subsequently, the spatial distribution rules of the collectors were studied by analyzing the spatial proportion, dispersion area, and uniformity and intensiveness of the collectors. It is an intuitive approach and a novel analysis method for the operating state of dust scrubber. The results show that the spatial distribution of the collectors could be better reflected by image processing methods. The dispersion area of the collectors expanded with an increase in the airflow velocity. When the initial liquid level (ILL) was higher, the collectors expanded in an approximate circular shape, and when the ILL was lower the collectors expanded in an approximate sector shape. In general, the variation trend in the spatial proportion enhanced with an increase in ILL and airflow velocity, which is consistent with the uniformity of collectors. When the liquid level was 0-20 mm and the airflow velocity was greater than 6.5 m/sec, the spatial proportion and uniformity of the collectors reached the highest degree. However, the growth rate of the spatial proportion and uniformity of the collectors slowed down and even led to negative growth when the ILL was lower and the airflow velocity was higher. The intensiveness of the collectors was great when the ILL was higher, which was free from the apparent influence of the airflow velocity and the ILL. However, when the ILL was lower, the intensiveness of the collectors was poor, intensifying as the airflow velocity and ILL increased. When the liquid level was -5-10 mm and the airflow velocity was greater than 8 m/sec, the intensiveness of the collectors reached the highest degree, indicating that a liquid level greater than 0 mm and a higher airflow velocity improved the spatial distribution of the collectors. Implications: This paper focuses on the spatial distribution of the collectors in dust scrubber. N-Fluorobenzenesulfonimide (NFSI) was identified for the first time as a highly effective Ag(i)-catalyst attenuator in the annulation of a tryptamine-derived ynesulfonamide to azepino[4,5-b]indole derivatives. Substrate tolerances were examined thoroughly and a plausible mechanism was proposed. The interaction between the Ag(i) catalyst and NFSI was probed by density functional theory (DFT) calculations. This study provided a highly efficient method to synthesize azepino[4,5-b]indole derivatives.

In order to address the bottleneck problem of low fine-particle removal efficiency of self-excited dust scrubbers, this paper is focused on the influence of the intermittent gas-liquid two-phase flow on the mesoscale behavior of collector aggregations. The latter is investigated by the application of high-speed dynamic image technology to the self-excited dust scrubber experimental setup. The real-time-scale monitoring of the dust removal process is provided to clarify its operating mechanism at the mesoscale level. The results obtained show that particulate capturing in self-excited dust scrubber is provided by liquid droplets, liquid films/curtains, bubbles, and their aggregations. Complex spatial and temporal structures are intrinsic to each kind of collector morphology, and these are considered as the major factors controlling the dust removal mechanism of self-excited dust scrubbers. For the specific parameters of gas-liquid two-phase flow under study, the evolution patterns of particular collectors reflect the intrinsic, intermittent, and complex characteristics of the temporal structure. The intermittent initiation of the collector and the air hole formation-collapse cyclic processes provide time and space for the fine dust to escape from being trapped by the collectors. The above mesoscale experimental data provide more insight into the factors reducing the dust removal efficiency of self-excited dust scrubbers. IMPLICATIONS: This paper focuses on the reconsideration of the capturer aggregations of self-excited dust scrubbers from the mesoscale. Complex structures in time and space scales exist in each kind of capturer morphology. With changes of operating parameters, the morphology and spatial distributions of capturers diversely change. The change of the capturer over time presents remarkable, intermittent, and complex characteristics of the temporal structure.

In Atlantic salmon (Salmo salar L.), and also in other fish species, certain plant protein ingredients can increase fecal water content creating a diarrhea-like condition which may impair gut function and reduce fish growth. The present study aimed to strengthen understanding of the underlying mechanisms by observing effects of various alternative plant protein sources when replacing fish meal on expression of genes encoding proteins playing key roles in regulation of water transport across the mucosa of the distal intestine (DI). A 48-day feeding trial was conducted with five diets: A reference diet (FM) in which fish meal (72%) was the only protein source; Diet SBMWG with a mix of soybean meal (30%) and wheat gluten (22%); Diet SPCPM with a mix of soy protein concentrate (30%) and poultry meal (6%); Diet GMWG with guar meal (30%) and wheat gluten (14.5%); Diet PM with 58% poultry meal. Compared to fish fed the FM reference diet, fish fed the soybean meal containing diet (SBMWG) showed signs of enteritis in the DI, increased fecal water content of DI chyme and higher plasma osmolality. Altered DI expression of a battery of genes encoding aquaporins, ion transporters, tight junction and adherens junction proteins suggested reduced transcellular transport of water as well as a tightening of the junction barrier in fish fed the SBMWG diet, which may explain the observed higher fecal water content and plasma osmolality. DI structure was not altered for fish fed the other experimental diets but alterations in target gene expression and fecal water content were observed, indicating that alterations in water transport components may take place without clear effects on intestinal structure.

Nickel complexes bearing amine-imine ligands with various backbone substituents were synthesized and employed as ethylene polymerization catalysts on activation with Et2 AlCl. The substituent on the backbone carbon atom of the amine moiety is decisive for the living nature of ethylene polymerization. A bulky amine-imine nickel precursor with a tert-butyl group on the carbon atom of the amine group can polymerize ethylene in a living fashion at an elevated temperature of 65 °C, which is the highest temperature of living polymerization of ethylene with late transition-metal catalysts. The wide applicable temperature range for living polymerization and sensitivity of the branch structure of the polyethylene to temperature enable precise synthesis of di- and triblock polyethylenes featuring different branched segments by sequential tuning of the polymerization temperature.

A bulky amine-imine nickel complex containing two 2,6-diisopropyl substituents after activation with MMAO or Et(2)AlCl can polymerize ethylene in a living fashion over a period of 120 minutes at room temperature or above.
